lncRNA - Long Non-coding RNAs02:39

lncRNA - Long Non-coding RNAs

8.4K
In humans, more than 80% of the genome gets transcribed. However, only around 2% of the genome codes for proteins. The remaining part produces non-coding RNAs which includes ribosomal RNAs, transfer RNAs, telomerase RNAs, and regulatory RNAs, among other types. A large number of regulatory non-coding RNAs have been classified into two groups depending upon their length – small non-coding RNAs, such as microRNA, which are less than 200 nucleotides in length, and long non-coding RNA...

Abnormal Proliferation02:23

Abnormal Proliferation

4.4K
Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. In such situations, cell division persists in the same excessive manner even after cessation of the stimuli, leading to persistent tumors. The tumor arises from the damaged cells that replicate to pass the damage to the...

Tumor Progression02:07

Tumor Progression

6.2K
Tumor progression is a phenomenon where the pre-formed tumor acquires successive mutations to become clinically more aggressive and malignant. In the 1950s, Foulds first described the stepwise progression of cancer cells through successive stages.
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...

mTOR Signaling and Cancer Progression03:03

mTOR Signaling and Cancer Progression

3.7K
The mammalian target of rapamycin or mTOR protein was discovered in 1994 due to its direct interaction with rapamycin. The protein gets its name from a yeast homolog called TOR. The mTOR protein complex in mammalian cells plays a major role in balancing anabolic processes such as the synthesis of proteins, lipids, and nucleotides and catabolic processes, such as autophagy in response to environmental cues, such as availability of nutrients and growth factors.
The mTOR pathway or the...

The Nucleolus02:55

The Nucleolus

8.6K
The nucleolus is the most prominent substructure of the nucleus. When it was first discovered, it was considered to be an isolated organelle that forms fibrils and granules. In 1931, the relationship between the nucleolus and chromosomes was first described by Heitz. He observed that the appearance and size of nucleolus varies depending on the stage of the cell cycle. He also noticed constricted regions on different chromosomes clustered together at definite cell cycle stages. These regions,...

Epigenetic Regulation01:37

Epigenetic Regulation

3.0K
Epigenetic changes alter the physical structure of the DNA without changing the genetic sequence and often regulate whether genes are turned on or off. This regulation ensures that each cell produces only proteins necessary for its function. For example, proteins that promote bone growth are not produced in muscle cells. Epigenetic mechanisms play an essential role in healthy development. Conversely, precisely regulated epigenetic mechanisms are disrupted in diseases like cancer.
X-chromosome...

  • Molecular Biology
  • Bioinformatics

Background:

  • hnRNPA3 is overexpressed in various cancers, including hepatocellular carcinoma (HCC).
  • The precise role and mechanisms of hnRNPA3 in HCC remain largely unelucidated.

Purpose of the Study:

  • To comprehensively analyze the function and potential mechanisms of hnRNPA3 in HCC using bioinformatics and experimental approaches.
  • To investigate hnRNPA3 as a potential diagnostic and prognostic biomarker for HCC.

Main Methods:

  • Utilized TCGA-LIHC dataset for pan-cancer expression, DNA methylation, ceRNA network, immune infiltration, and immune checkpoint gene analysis.
  • Employed logistic regression, ROC curves, Kaplan-Meier analysis, and nomogram modeling for prognostic evaluation.
  • Conducted in vitro experiments including qRT-PCR, Western blot, cell proliferation/migration assays, and RNA-seq.

Main Results:

  • hnRNPA3 expression was significantly elevated in HCC, correlating with clinical parameters and predicting survival.
  • Associations between hnRNPA3, DNA methylation, and a ceRNA network involving hsa-miR-22-3p were identified.
