Kainate receptors (KARs) undergo unique conformational changes during desensitization, unlike other ionotropic glutamate receptors (iGluRs). Structural and functional data reveal that KAR ligand-binding domain rotation is crucial for complete channel closure and distinct desensitization mechanisms.
