    Area of Science:

    • Microbiology
    • Immunology
    • Vaccine Development

    Background:

    • Pseudomonas aeruginosa is a significant opportunistic pathogen.
    • Developing effective vaccines against diverse serotypes remains a challenge.

    Purpose of the Study:

    • To prepare and characterize a new polyvalent Pseudomonas vaccine.
    • To evaluate the immunogenic properties and protective efficacy of the vaccine.

    Main Methods:

    • Developed novel cultural methods for enhanced immunogen production in bacterial cell walls.
    • Utilized mild extraction techniques to preserve immunogen integrity.
    • Formulated a polyvalent vaccine comprising 16 component vaccines (lipid-protein-carbohydrate complexes).

    Main Results:

    • Single injections of component vaccines conferred protection against homologous serotypes in mice within 3 days.
    • The polyvalent vaccine demonstrated protection against multiple strains of all 16 serotypes.
    • Extraction methods preserved the immunogenicity of the extracted components.

    Conclusions:

    • The developed polyvalent Pseudomonas vaccine is effective in inducing protection against multiple serotypes.
    • The novel preparation and extraction methods are suitable for producing immunogenic bacterial vaccines.
