Association of GCH1 polymorphism with Parkinson disease susceptibility: A meta-analysis

Background:
Guanosine triphosphate cyclohydrolase 1 (GCH1) gene rs11158026 polymorphism was suggested to be associated with Parkinson disease (PD) risk; however, the conclusions remain controversial. Thus, we performed a meta-analysis to investigate the association between GCH1 rs11158026 polymorphism and PD susceptibility.
Methods:
The literature was searched from electronic databases such as PubMed, Cochrane Library, Embase, Web of Science, and Chinese national knowledge infrastructure until March 1, 2024. The odds ratio (ORs) and 95% confidence intervals (95% CIs) were used to assess the relationship between the GCH1 rs11158026 polymorphism and PD susceptibility. Data analysis was processed by Stata 12.0 software.
Results:
Six articles with 4091 cases and 4863 controls were included in this meta-analysis. There was no significant association between GCH1 rs11158026 polymorphism and PD risk in all genetic models. In subgroup analysis by ethnicity showed a strong association in Caucasians (TT vs CT + CC: OR = 0.70, 95% CI = 0.55-0.90, P = .005; TT vs CC: OR = 0.69, 95% CI = 0.52-0.92, P = .011; T vs C: OR = 0.85, 95% CI = 0.74-0.98, P = .025).
Conclusion:
Our findings suggested that GCH1 rs11158026 polymorphism may be associated with PD susceptibility in Caucasians, but the results should be interpreted with caution due to insufficient evidence.
