The Tumor Microenvironment02:17

The Tumor Microenvironment

6.4K
Every normal cell or tissue is embedded in a complex local environment called stroma, consisting of different cell types, a basal membrane, and blood vessels. As normal cells mutate and develop into cancer cells, their local environment also changes to allow cancer progression. The tumor microenvironment (TME) consists of a complex cellular matrix of stromal cells and the developing tumor. The cross-talk between cancer cells and surrounding stromal cells is critical to disrupt normal tissue...

Tumor Immunotherapy01:27

Tumor Immunotherapy

398
Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Summary

Teashirt zinc finger homeobox 3 (TSHZ3) acts as a tumor suppressor in lung adenocarcinoma (LUAD). Lower TSHZ3 expression correlates with poor prognosis, while its overexpression inhibits cancer progression and metastasis.

Area of Science:

  • Oncology
  • Molecular Biology
  • Immunology

Background:

  • Teashirt zinc finger homeobox 3 (TSHZ3) is a transcription factor involved in cancer progression.
  • Its specific role and expression in lung adenocarcinoma (LUAD) are not well understood.

Purpose of the Study:

  • To investigate TSHZ3 expression and prognostic value in LUAD.
  • To elucidate the functional mechanisms of TSHZ3 in LUAD progression.

Main Methods:

  • Bioinformatics analysis of prognostic data and functional prediction.
  • In vitro and in vivo cellular and animal experiments.
  • Gene Ontology (GO) and pathway analyses, immune infiltration analysis.

Main Results:

  • TSHZ3 expression is significantly lower in LUAD tissues compared to normal tissues.
  • High TSHZ3 expression is associated with improved overall survival in LUAD patients.
  • TSHZ3 overexpression inhibits LUAD cell migration, invasion, and epithelial-mesenchymal transition (EMT), promoting apoptosis via M1 macrophage recruitment through the CCL2/CCR2 pathway.

Conclusions:

  • TSHZ3 functions as a tumor suppressor in LUAD.
  • TSHZ3 inhibits LUAD progression by suppressing EMT and metastasis.
  • TSHZ3 induces apoptosis by recruiting M1 macrophages, highlighting its potential as a therapeutic target.
