Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. Summary

Leucine-rich alpha-2-glycoprotein-1 (LRG1) impairs developing retinal blood vessels, reducing pericyte coverage and vessel maturation. This angiopathic factor highlights potential LRG1-blocking therapies for vascular diseases.

Background:

  • Leucine-rich alpha-2-glycoprotein-1 (LRG1) is implicated in aberrant neovascularization.
  • LRG1's role in developmental angiogenesis is not well-understood.

Purpose of the Study:

  • To investigate the effect of LRG1 on developing retinal blood vessels.
  • To determine if LRG1 possesses vasculopathic properties during vascular development.

Main Methods:

  • Exogenous LRG1 (recombinant protein or expression vector) was injected into mouse retinas during development.
  • Effects on pericyte coverage, NG2 expression, collagen IV, junctions, vessel calibre, and vascular density were assessed.
  • Blood-retinal barrier permeability and transcytosis were evaluated.

Main Results:

  • Exogenous LRG1 reduced pericyte coverage and NG2 expression.
  • Diminished collagen IV sheathing, fewer cell junctions, and reduced vessel calibre/density were observed.
  • Increased blood-retinal barrier permeability and transcytosis were noted.

Conclusions:

  • Exogenous LRG1 interferes with retinal vessel maturation, promoting a dysfunctional phenotype.
  • These findings establish LRG1 as an angiopathic factor.
  • Blocking LRG1 may offer therapeutic potential for angiogenic and vascular remodeling diseases.
