A novel mutation in GBPL2 suppresses defects in plant immunity related to CONSTITUTIVE EXPRESSION OF PR GENES 5 (CPR5). This suppressor mutation reveals that GBPL2 interacts with GBPL3 to regulate immune signaling downstream of CPR5.
Area of Science:
- Plant molecular biology
- Plant immunity and defense mechanisms
- Cellular transport and signaling
Background:
- Nuclear pore complex is crucial for immune signal transport.
- CONSTITUTIVE EXPRESSION OF PR GENES 5 (CPR5) and GUANYLATE-BINDING PROTEIN-LIKE 3 (GBPL3) are involved in plant immunity.
- The function of GBPL2 within the GUANYLATE-BINDING PROTEIN-LIKE (GBPL) family was previously unknown.
Purpose of the Study:
- To investigate the unknown role of GBPL2 in plant immunity.
- To identify genetic factors that modulate the CPR5 immune pathway.
- To elucidate the molecular mechanism of GBPL family proteins in plant defense.
Main Methods:
- Genetic screening to identify suppressors of the cpr5 mutant.
- Characterization of a gain-of-function mutation in GBPL2 (scpr23).
- Analysis of protein localization, epistatic interactions, and gene expression.
Main Results:
- A gain-of-function mutation in GBPL2 (scpr23) was identified, which fully suppresses the cpr5 mutant phenotype.
- The scpr23 mutation alters GBPL2 localization from the ER to the nuclear envelope by affecting a nuclear export signal (NES).
- Epistatic analysis confirmed that GBPL2's function in suppressing cpr5 is mediated through GBPL3, suggesting a GBPL2-GBPL3 signaling complex downstream of CPR5.
Conclusions:
- GBPL2, particularly through its scpr23 mutant form, plays a significant role in regulating plant immunity.
- The GBPL family proteins, including GBPL2 and GBPL3, form a signaling complex downstream of CPR5.
- GBPL2 acts as a plant ortholog to mammalian ATLASTIN-1, highlighting conserved functions in membrane dynamics and immunity.
