Summary

Polycystin 1 (PC1) and polycystin 2 (PC2) genes are upregulated in astrocytomas, particularly glioblastomas. Their increased expression correlates with poorer prognosis in glioma patients, suggesting a role in tumor development.

Background:

  • Polycystin 1 (PC1) and polycystin 2 (PC2) are TRP channel proteins encoded by PKD1 and PKD2 genes.
  • While known for autosomal dominant polycystic kidney disease, polycystins are implicated in solid tumor pathogenesis.
  • Expression of PKD1 and PKD2 in human astrocytomas remains largely uncharacterized.

Purpose of the Study:

  • To investigate PKD1 and PKD2 gene expression in astrocytic tumors.
  • To correlate PC1 and PC2 protein expression with clinicopathological features of astrocytomas.
  • To assess the relationship between polycystin expression and tumor grade, patient age, and gender.

Main Methods:

  • Examined 70 astrocytoma cases (8 grade II, 12 grade III, 50 grade IV).
  • Quantified mRNA levels of PKD1 and PKD2 using qRT-PCR (ΔΔCt method).
  • Detected PC1 and PC2 protein expression via immunohistochemistry (H-score system).

Main Results:

  • PKD1 and PKD2 mRNA levels, along with PC1 and PC2 proteins, were significantly elevated in astrocytomas compared to normal brain tissue.
  • Glioblastomas (grade IV) showed the highest increase in PKD1, PKD2, PC1, and PC2 expression.
  • PC1 expression correlated with patient age, and both PC1 and PC2 showed correlations with p53 immunoexpression in specific grades. PKD1 and PKD2 expression negatively correlated with glioma prognosis.

Conclusions:

  • Polycystins (PC1 and PC2) are potentially involved in astrocytoma pathogenesis.
  • Elevated polycystin expression in astrocytomas, especially glioblastomas, suggests a role in tumor progression.
  • Further research is needed to elucidate the precise mechanisms of polycystin involvement in astrocytoma development.
