This study identifies enhancer-associated long noncoding RNA-ITGA2 (LncRNA-ITGA2) as a key driver of vascular smooth muscle cell proliferation and migration. LncRNA-ITGA2 promotes vascular remodeling by interacting with NONO to enhance ITGA2 gene expression.
Area of Science:
- Vascular Biology
- Epigenetics
- Molecular Mechanisms of Disease
Background:
- Vascular smooth muscle cell (VSMC) proliferation and migration are critical in vascular remodeling.
- Enhancer-associated long noncoding RNAs (elncRNAs) are implicated in cell fate, but their role in VSMC dysfunction is unclear.
Purpose of the Study:
- To identify and characterize novel elncRNAs involved in VSMC dysfunction.
- To elucidate the regulatory mechanisms of elncRNAs in vascular remodeling.
Main Methods:
- Multiomics profiling (CUT&Tag, Hi-C, microarray) to identify LncRNA-ITGA2.
- In vivo carotid artery wire injury model and in vitro VSMC culture.
- RNA sequencing, CRISPR-Cas9, ChIP-seq, ChIRP, and RIP to investigate mechanisms.
Main Results:
- LncRNA-ITGA2 is upregulated in proliferative VSMCs and atherosclerotic tissues.
- Overexpression of LncRNA-ITGA2 promotes VSMC proliferation, migration, and neointimal hyperplasia.
- LncRNA-ITGA2 enhances ITGA2 expression by facilitating enhancer-promoter interaction and H3K27 acetylation, dependent on NONO.
Conclusions:
- LncRNA-ITGA2 promotes VSMC proliferation and migration via a NONO-dependent mechanism, linking enhancer-ITGA2 and promoter-ITGA2.
- This elncRNA is a potential diagnostic and therapeutic target for proliferative vascular diseases.
