Tumor Immunotherapy01:27

Tumor Immunotherapy

401
Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Combination Therapies and Personalized Medicine

4.8K
Combining two or more treatment methods increases the life span of cancer patients while reducing damage to vital organs or tissue from the overuse of a single treatment. Combination therapy also targets different cancer-inducing pathways, thus reducing the chances of developing resistance to treatment.
The combination of the drug acetazolamide and sulforaphane is a good example of combination therapy to treat cancer. The cells in the interior of a large tumor often die due to the hypoxic and...

Area of Science:

  • Oncology
  • Immunotherapy
  • Gastroenterology

Background:

  • Colorectal cancer (CRC) presents a significant global health burden.
  • Most CRC patients have microsatellite stable (MSS) and proficient mismatch repair (pMMR) tumors.
  • These MSS/pMMR tumors are typically resistant to immune checkpoint inhibitors (ICIs).

Purpose of the Study:

  • To explore the potential of combining cetuximab with ICIs for MSS/pMMR CRC.
  • To investigate if this combination can overcome intrinsic resistance to immunotherapy.
  • To evaluate the synergistic effects of cetuximab and ICIs in MSS/pMMR CRC.

Main Methods:

  • Review of preclinical and clinical data on cetuximab and ICI combinations.
  • Analysis of the immunomodulatory properties of cetuximab.
  • Assessment of complementary therapeutic mechanisms.

Main Results:

  • Cetuximab, an EGFR-targeting antibody, possesses immunomodulatory properties.
  • Combination therapy may synergistically enhance response rates and durability.
  • Preclinical and early clinical data suggest promising outcomes.

Conclusions:

  • Combining cetuximab with ICIs is a potential strategy for MSS/pMMR CRC.
  • Further large-scale, randomized phase III trials are essential.
  • Ongoing trials will clarify the role of this combination in personalized immunotherapy for CRC.
