Area of Science:

  • Materials Science
  • Nanotechnology
  • Immunology

Background:

  • Rheumatoid arthritis (RA) is a debilitating autoimmune joint disease.
  • Neutrophils are key drivers of RA inflammation, presenting therapeutic targets.

Purpose of the Study:

  • To develop and evaluate iron-doped carbon dots (MM-CDs) as a novel RA therapy.
  • To investigate the mechanism of MM-CDs in mitigating neutrophil-mediated inflammation.

Main Methods:

  • MM-CDs synthesized using magnetite and medicated leaven.
  • Therapeutic efficacy assessed in a collagen-induced arthritis mouse model.
  • Analysis of neutrophil extracellular trap (NETosis) and related markers (MPO, Cit-H3, PAD-4).
  • Evaluation of autophagy (LC3B, Beclin-1) and apoptosis (caspase-3, PARP, Bcl-2) pathways.

Main Results:

  • MM-CDs significantly alleviated RA symptoms in mice.
  • Inhibition of NETosis and downregulation of MPO, Cit-H3, and PAD-4 observed.
  • Modulation of autophagy and promotion of neutrophil apoptosis were confirmed.

Conclusions:

  • MM-CDs represent a promising therapeutic strategy for rheumatoid arthritis.
  • The treatment demonstrated no observable liver or kidney toxicity in the RA mouse model.
  • MM-CDs show potential for safe clinical translation in RA treatment.
