Ovarian Cycle01:27

Ovarian Cycle

1.1K
The menstrual cycle includes a critical component known as the ovarian cycle, which undergoes two main phases each month—the follicular phase and the luteal phase. The follicular phase is variable and averaging around 14 days. Ovulation, triggered by a surge in luteinizing hormone (LH), marks the transition between the two phases. The second phase, the luteal phase, is relatively consistent, lasting approximately 14 days, and is marked by the activity of the corpus luteum. While a cycle...

Cleavage and Blastulation01:33

Cleavage and Blastulation

44.9K
After a large-single-celled zygote is produced via fertilization, the process of cleavage occurs while zygotes travel through the uterine tube. Cleavage is a mitotic cell division that does not result in growth. With each round of successive cell division, daughter cells get increasingly smaller.

Gonadal and Placental Hormones01:24

Gonadal and Placental Hormones

1.4K
The gonads, namely the testes in males and the ovaries in females, are pivotal in producing gonadal hormones that orchestrate the intricate processes of sexual development and reproduction.
In males, testosterone is the primary gonadal androgen. It plays a central role in the maturation of male reproductive organs — the penis and testes. Additionally, testosterone is instrumental in the development of secondary sexual characteristics — a deep voice as well as facial and pubic hair...

Proliferative Phase01:20

Proliferative Phase

411
The proliferative phase typically occurs after menstruation and lasts between 6 to 13 days in a standard 28-day cycle. This phase involves the reconstruction of the endometrium, guided by estrogen produced by the developing ovarian follicle.
Notably, the stratum basale, the basal layer of the endometrium, including the basal parts of the uterine glands, remains unaffected by menstruation. Stem cells in this layer undergo mitosis, regenerating the stratum functionalis and thickening the...

Summary

Embryo-derived human chorionic gonadotropin (hCG) stimulates uterine PGF2α production, which then promotes decidualization crucial for pregnancy. This pathway involves epithelial cells and key signaling molecules like EREG and HB-EGF.

Area of Science:

  • Reproductive Biology
  • Endocrinology
  • Cell Biology

Background:

  • Decidualization of endometrial stromal cells is vital for successful pregnancy.
  • Dysfunctional decidualization is linked to implantation failure and recurrent miscarriage.
  • The role of prostaglandin F2α (PGF2α) in embryo implantation and decidualization remains unclear.

Purpose of the Study:

  • To investigate the role of human chorionic gonadotropin (hCG) in stimulating PGF2α production in the uterus.
  • To elucidate the signaling pathway by which hCG influences decidualization.
  • To determine if PGF2α is involved in embryo implantation and human in vitro decidualization.

Main Methods:

  • Human endometrial epithelial cells were treated with hCG.
  • Levels of PGF2α synthesis-related enzymes (cPLA2, COX-1, COX-2, AKR1C3) and PGF2α secretion were measured.
  • The effects of PGF2α, EREG, and HB-EGF on IGFBP1 expression (a marker of decidualization) were assessed in vitro.

Main Results:

  • hCG stimulation led to increased PGF2α synthesis-related enzymes and PGF2α secretion by human endometrial epithelial cells.
  • Epithelial PGF2α induced the secretion of EREG and HB-EGF via ADAM17 activation.
  • EREG, HB-EGF, and PGF2α all significantly increased IGFBP1 levels, indicating promotion of in vitro decidualization.

Conclusions:

  • hCG, an embryo-derived protein, stimulates uterine epithelial cells to produce PGF2α.
  • This PGF2α then acts through EREG and HB-EGF to promote human in vitro decidualization.
  • The findings reveal a novel pathway linking hCG to decidualization, crucial for early pregnancy.
