    Summary

    Pentobarbital causes drinking in rats, but this effect does not depend on the kidneys or renin release. This suggests central nervous system mechanisms are involved in pentobarbital-induced thirst.

    Background:

    • Pentobarbital induces drinking in rats.
    • Pentobarbital is a known renin releaser.
    • Renin plays a role in thirst regulation.

    Purpose of the Study:

    • To investigate if pentobarbital-induced drinking depends on a kidney-released substance (renal dipsogen).
    • To determine if blocking renin release via nephrectomy affects pentobarbital's effect on drinking.

    Main Methods:

    • Rats underwent either sham surgery or nephrectomy (kidney removal).
    • Animals were injected with pentobarbital sodium or a vehicle control.
    • Water intake was measured 60 minutes post-injection.

    Main Results:

    • Pentobarbital induced significant drinking in both sham-operated and nephrectomized rats.
    • No significant difference in water intake was observed between the two groups.
    • These findings indicate pentobarbital-induced drinking is independent of renal renin release.

    Conclusions:

    • Pentobarbital-induced drinking is not mediated by increased plasma renin activity.
    • Central nervous system mechanisms likely underlie the dipsogenic response to pentobarbital.
