Chromatin Immunoprecipitation- ChIP

11.3K
Chromatin immunoprecipitation, or ChIP, is an antibody-based technique used to identify sites on DNA that bind to transcription factors of interest or histone proteins. It also helps determine the type of histone modifications such as acetylation, phosphorylation, or methylation.
Types of ChIP
ChIP can be divided into two types - X-ChIP and N-ChIP. X-ChIP involves in vivo cross-linking of histones and regulatory proteins to DNA, fragmenting the DNA by sonication, and isolating the protein-DNA...

Area of Science:

  • Cytopathology
  • Oncology
  • Computational Biology

Background:

  • Previous development of the chromatin homology profile (CHP) method for lung cancer histology differentiation in respiratory cytology.
  • Challenges with the 100× objective lens, including field of view and data volume, limited the prior CHP method's application.
  • This study aimed to address these limitations by evaluating CHP accuracy with a 40× objective lens.

Purpose of the Study:

  • To investigate the accuracy of the chromatin homology profile (CHP) method for differentiating lung cancer histological types using 40× objective microscopy.
  • To determine if the CHP method, when applied to images from a 40× objective, can effectively distinguish between benign and malignant respiratory cytology cases.
  • To assess the feasibility of using the CHP method as a diagnostic support system in respiratory cytology.

Main Methods:

  • Selection of 35 cases of lung cancer and benign tissue.
  • Imaging of cancer cells and ciliated columnar epithelial cells using a 40× objective lens on a KEYENCE microscopy system.
  • Analysis of chromatin contact (b1MAX) and chromatin density (b1MAX/nuclear area) using the Cell Checker application.

Main Results:

  • The b1MAX value showed significant differences between small cell carcinoma and non-small cell carcinoma.
  • b1MAX/nuclear area values revealed significant differences between adenocarcinoma and squamous cell carcinoma.
  • Significant differences in both b1MAX and b1MAX/nuclear area values were observed between benign and malignant cases, mirroring results from the 100× objective.

Conclusions:

  • The chromatin homology profile (CHP) method, using 40× objective images, successfully overcomes previous technical challenges.
  • The CHP method demonstrates accuracy in differentiating benign from malignant respiratory cytology cases.
  • The CHP method is a viable tool for a diagnostic support system in respiratory cytology.
