Blood metabolomic and Crohn's disease: A Mendelian randomization study

Crohn's disease, a type of inflammatory bowel disease, is marked by chronic inflammation of the gastrointestinal tract, leading to symptoms like abdominal pain, severe diarrhea, fatigue, weight loss, and malnutrition. The pathophysiology of Crohn's disease is complex and not fully understood, with recent studies suggesting a potential role for metabolic dysregulation in its onset and progression. The exact causal relationships, however, remain elusive. In this study, a 2-sample Mendelian randomization (MR) approach was used to examine the potential causal relationship between 1091 blood metabolites and 309 metabolite ratios, and the occurrence of Crohn's disease. Utilizing genetic variants as instrumental variables, the analysis was based on data sourced from genome-wide association studies databases for a population of European descent. The selection of instrumental variables was based on strict criteria, and the study utilized multiple statistical methods such as inverse variance weighting, weighted median, and mode-based techniques. The MR analysis identified 11 metabolites with potential causal links to Crohn's disease. Out of these, 7 metabolites showed protective effects, whereas 4 were linked to an elevated risk of the condition. The results were consistent among various MR methods, suggesting a strong correlation. This research emphasizes the capacity of metabolomics for comprehending Crohn's disease. The identification of specific metabolites associated with the condition provides new insights into its pathophysiology and opens avenues for future research into targeted treatment strategies.
