    Purpose of the Study:

    • To investigate rat Tg kinetics after separating 19S Tg from its derivatives.
    • To elucidate the influence of circulating Tg derivatives on kinetic study results.

    Main Methods:

    • Rats were injected with 125I; radiolabeled Tg was purified.
    • Plasma samples were fractionated using Sephacryl S-300 gel-filtration.
    • Sucrose density gradient confirmed 99% of in vivo radiolabeled Tg as 19S.

    Main Results:

    • A second radioactive peak (MW 60,000-70,000) appeared within 6 hours post-injection.
    • This second peak contained Tg-related radioactivity, affecting TCA precipitation and butanol extraction.
    • The presence of this peak can interfere with Tg kinetic studies.

    Conclusions:

    • Circulating rat Tg derivatives significantly affect the accuracy of kinetic studies.
    • Separation of 19S Tg from derivatives is crucial for precise half-life determination.
    • Standard kinetic study methods (TCA precipitation, RIA) may yield inaccurate results due to these derivatives.
