A new asymmetric Suzuki-Miyaura cross-coupling (ASMC) reaction enables direct synthesis of chiral biaryl monophosphines. This method overcomes challenges with phosphorus(III) compounds, yielding effective ligands for catalysis.
Area of Science:
- Organic Chemistry
- Catalysis
- Asymmetric Synthesis
Background:
- The Suzuki-Miyaura cross-coupling (SMC) is vital for biaryl synthesis.
- Enantioselective SMC typically requires external chiral ligands.
- Phosphorus(III) compounds pose challenges due to strong metal coordination.
Purpose of the Study:
- To develop a P(III)-compatible asymmetric Suzuki-Miyaura cross-coupling (P-ASMC) reaction.
- To enable one-step synthesis of axially chiral biaryl monophosphines.
- To identify key factors for reaction efficiency and enantioselectivity.
Main Methods:
- Utilized a palladium catalyst with achiral and chiral phosphine ligands.
- Coupled ortho-bromo-substituted arylphosphines with aryl boronate esters.
- Conducted experimental and computational studies to elucidate reaction mechanisms.
Main Results:
- Achieved high reactivity and enantioselectivity in the P-ASMC reaction.
- Successfully synthesized axially chiral biaryl monophosphines in one step.
- Demonstrated the efficacy of the synthesized atropisomers as ligands in various metal catalysis.
Conclusions:
- The P-ASMC reaction provides a novel route to valuable chiral phosphine ligands.
- The developed method overcomes previous limitations in coupling phosphorus(III) compounds.
- The synthesized ligands show broad applicability in enantioselective catalysis.

Aldehydes and Ketones to Alkenes: Wittig Reaction Mechanism
The reaction begins with the nucleophilic addition between a phosphorus ylide and the carbonyl compound. Due to its carbanionic character, phosphorus ylide acts as a strong nucleophile and attacks the electrophilic carbonyl group. This generates a charge-separated dipolar intermediate called betaine. The negatively charged oxygen atom and...
