Area of Science:

  • Oncology
  • Immunology
  • Genetics

Background:

  • Clear cell renal cell carcinoma (ccRCC) is an aggressive kidney cancer subtype.
  • The roles of lactate metabolism and immune-related genes (LMRGs) in ccRCC prognosis and immune microenvironment are not well understood.
  • No prior studies have investigated the prognostic impact of LMRGs in ccRCC.

Purpose of the Study:

  • To explore the potential impact of LMRGs on ccRCC prognosis.
  • To investigate the relationship between LMRGs and the tumor immune microenvironment in ccRCC.
  • To identify potential biomarkers for ccRCC prognosis based on LMRGs.

Main Methods:

  • Transcriptomic analysis using TCGA and GEO databases.
  • Non-negative matrix factorization (NMF) for ccRCC sample subtyping.
  • LASSO regression and Cox analysis for core prognostic gene identification.
  • Kaplan-Meier survival and CIBERSORT/ESTIMATE analyses for survival and immune infiltration assessment.

Main Results:

  • ccRCC samples were classified into two subtypes by NMF, with Cluster 2 showing better prognosis.
  • Five key genes (STAT2, PDGFRL, APLNR, PRKCQ, THRB) were identified and used to build a prognostic model.
  • High-risk group demonstrated lower survival rates, higher immune cell infiltration, and TMB synergy.
  • Low-risk group was enriched for metabolism-related pathways.

Conclusions:

  • LMRGs are significant for prognostic evaluation in ccRCC.
  • The identified core genes hold potential as prognostic biomarkers for ccRCC.
  • Understanding LMRG roles can improve ccRCC prognosis and treatment strategies.
