Dosage Regimen: Fixed Dose01:01

Dosage Regimen: Fixed Dose

1.9K
Fixed-dose regimens are a common approach to administer drugs to achieve and maintain desired levels of the drug in the body. In this dosing strategy, a specific amount of medication is given at regular intervals, often multiple times a day, to ensure a consistent drug concentration in the bloodstream.
Fixed-dose regimens can be used for various routes of administration, including intravenous (IV) injections and oral medications. For IV administration, a predetermined amount of the drug is...

Effective preventive measures for coronary artery disease (CAD) focus on controlling modifiable risk factors, including cholesterol abnormalities and lifestyle changes.Cholesterol ManagementFirst, the Mediterranean diet and the American Heart Association advocate for maintaining low-density lipoprotein (LDL) cholesterol levels below 100 mg/dL, with a more stringent recommendation of below 70 mg/dL for individuals at high risk. LDL cholesterol, often termed "bad cholesterol," can lead to the...

Atherosclerosis III: Management

2
Management of atherosclerosis involves an integrated strategy encompassing pharmacological treatment, surgical interventions, lifestyle changes, and nutrition therapy to address the multifactorial nature of the disease.Pharmacological TherapyA cornerstone of atherosclerosis management is the use of pharmacological agents. Statins, such as atorvastatin, are pivotal in inhibiting HMG-CoA reductase, an enzyme that catalyzes an initial step in cholesterol synthesis in the liver. This reduction in...

IntroductionThe management of angina requires a comprehensive approach that includes pharmacological therapies, medical procedures, and lifestyle modifications.Pharmacological TherapiesAntiplatelet agents, such as aspirin, clopidogrel, prasugrel, and ticagrelor, play a pivotal role in preventing thrombus formation in patients with angina. These medications inhibit platelet aggregation and reduce the likelihood of myocardial infarction and other cardiovascular events.Anticoagulants, including...

Antiplatelet Drugs: Prostaglandin Synthesis, P2Y12 and Glycoprotein IIb/IIIa Inhibitors

Antiplatelet drugs emerge as frontline defenders against the insidious threat of thromboembolic diseases, where abnormal clots obstruct vital blood vessels. These drugs stand as bulwarks, inhibiting platelet aggregation and clot formation, thereby mitigating the risk of life-threatening conditions like myocardial infarction, coronary artery disease, and thrombotic strokes.
Prostaglandin synthesis inhibitors, exemplified by the widely known aspirin, wield their power by irreversibly acetylating...

Interprofessional care for coronary artery disease includes pharmacological therapy and revascularization procedures.Pharmacological therapy for Coronary Artery Disease (CAD) aims to manage symptoms, prevent complications, and improve patient outcomes through various classes of medications:Antiplatelet Agents:Aspirin and Clopidogrel: These medications inhibit platelet aggregation, preventing blood clots, which is crucial for avoiding heart attacks and strokes. Doctors often prescribe these...

Summary

Polypill intervention significantly reduces myocardial infarction (MI) and major adverse cardiovascular events (MACE) incidence. This fixed-dose combination also lowers blood pressure and LDL cholesterol levels, improving cardiovascular outcomes.

Area of Science:

  • Cardiovascular Medicine
  • Pharmacology
  • Public Health

Background:

  • Myocardial infarction (MI) remains a leading cause of cardiovascular disease (CVD) mortality.
  • Existing treatments for MI and related risk factors often yield unsatisfactory outcomes.
  • Polypill therapy offers a potential strategy for comprehensive cardiovascular risk reduction.

Purpose of the Study:

  • To evaluate the efficacy of polypill intervention in preventing MI.
  • To assess the impact of polypills on key cardiovascular risk factors, including blood pressure and blood lipids.
  • To analyze the effect of polypills on major adverse cardiovascular events (MACE) and mortality.

Main Methods:

  • Systematic search and meta-analysis of individual participant data from randomized controlled trials (RCTs).
  • Comparison of polypill (fixed-dose combination) versus usual care in preventing MI.
  • Primary outcomes included MI, CVD mortality, MACE, and all-cause mortality; secondary outcomes included blood pressure and lipid levels.

Main Results:

  • Polypill use significantly reduced the incidence of MI (OR=0.76) and MACE (OR=0.79) compared to usual care.
  • Significant reductions were observed in systolic blood pressure (SMD=-0.13), diastolic blood pressure (SMD=-0.13), and LDL cholesterol (SMD=-0.21).
  • No statistically significant reductions were found for CVD mortality, all-cause mortality, HDL, or total cholesterol levels.

Conclusions:

  • Polypill intervention demonstrates high effectiveness in preventing MI and reducing MACE incidence.
  • Polypills contribute to lowering blood pressure and LDL cholesterol levels, addressing key cardiovascular risk factors.
  • The findings support polypill use as a valuable strategy in cardiovascular disease prevention.
