Prodrugs

2.5K
Prodrugs are a class of pharmaceutical compounds that undergo a biotransformation process within the body to be converted into a pharmacologically active drug. Prodrugs are designed to improve the therapeutic properties of the parent drug, such as enhancing bioavailability, increasing stability, or reducing toxicity. The concept of prodrugs revolves around modifying the chemical structure of the original drug to make it more effective or convenient for administration.

Targeted Cancer Therapies

7.5K
The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against...

Summary

Ligand-targeted drugs (LTDs) targeting prostate-specific membrane antigen (PSMA) show promise for metastatic castration-resistant prostate cancer. These PSMA-targeting LTDs deliver cytotoxic agents directly to cancer cells, outperforming traditional chemotherapy.

Area of Science:

  • Oncology
  • Pharmacology
  • Biochemistry

Background:

  • Prostate-specific membrane antigen (PSMA), also known as glutamate carboxypeptidase II (GCPII), is highly expressed in prostate cancer cells.
  • PSMA's specific expression profile makes it an attractive target for cancer therapy.
  • Ligand-targeted drugs (LTDs) offer a strategy to deliver cytotoxic agents specifically to PSMA-expressing tumors.

Purpose of the Study:

  • To review the structure and preclinical data of PSMA-targeting LTDs.
  • To discuss the clinical progress of promising PSMA-targeting LTDs for prostate cancer treatment.

Main Methods:

  • Review of existing literature on PSMA-targeting LTDs.
  • Analysis of preclinical in vitro and in vivo study results.
  • Evaluation of available clinical trial data for selected PSMA-targeting LTDs.

Main Results:

  • PSMA-targeting LTDs demonstrate high affinity and specificity for prostate cancer cells.
  • Preclinical studies show promising efficacy of these targeted agents.
  • Several PSMA-targeting LTDs are advancing through clinical trials with encouraging outcomes.

Conclusions:

  • PSMA-targeting LTDs represent a significant advancement in the treatment of metastatic castration-resistant prostate cancer.
  • These targeted therapies have the potential to improve efficacy and reduce systemic toxicity compared to conventional cytotoxic drugs.
