This study evaluated implicit solvent models for dacarbazine-cyclodextrin complexes. The Solvation Model based on Density (SMD) best matched experimental data for thermodynamic properties, particularly for HE-β-CD complexes.
Area of Science:
- Computational Chemistry
- Supramolecular Chemistry
Background:
- Dacarbazine is an anticancer drug.
- Cyclodextrins (CDs) are used to improve drug solubility and stability.
- Understanding drug-CD complexation is crucial for pharmaceutical development.
Purpose of the Study:
- To evaluate the influence of implicit solvent models on dacarbazine-cyclodextrin complex properties.
- To compare theoretical thermodynamic data with experimental results.
- To identify the most stable dacarbazine-cyclodextrin complex and the best-performing solvent model.
Main Methods:
- Computational modeling using four implicit solvent models: Polarizable Continuum Model (PCM), Conductor-like Polarizable Continuum Model (CPCM), Solvation Model based on Density (SMD), and Onsager model.
- Analysis of structural and thermodynamic properties of dacarbazine complexes with α-CD, HP-β-CD, and HE-β-CD.
- Comparison of theoretical results with experimental data from conductometric studies.
Main Results:
- All three cyclodextrins formed stable 1:1 inclusion complexes with dacarbazine.
- Complexes with HE-β-CD exhibited the highest thermodynamic stability.
- The choice of solvent model significantly impacted thermodynamic parameters but had a minor effect on structural properties.
- The SMD model showed the best agreement with experimental data, especially for Gibbs free energy of solvation.
Conclusions:
- Implicit solvent models significantly influence the calculated thermodynamic properties of dacarbazine-cyclodextrin complexes.
- The SMD model provides the most accurate theoretical prediction of these properties compared to experimental data.
- HE-β-CD is the most effective cyclodextrin for forming stable dacarbazine inclusion complexes.

Stability of Substituted Cyclohexanes
The two chair conformations of cyclohexanes undergo rapid interconversion at room temperature. Both forms have identical energies and stabilities, each comprising equal amounts of the equilibrium mixture. Replacing a hydrogen atom with a functional group makes the two conformations energetically non-equivalent.
For example, in...
Stability of Conjugated Dienes
A comparison of the enthalpies of hydrogenation of dienes reveals that conjugated dienes release less heat on hydrogenation, rendering them more stable than their nonconjugated analogs.
Conformations of Cyclohexane
The chair form is the most stable and derives its name from its resemblance to the “easy chair.” In the chair conformation, two carbon atoms are arranged out-of-plane — one above and one below, minimizing the torsional strain. In the chair form, the bond angle is very close to the ideal...
