A reaction based fluorescent sensor for the detection of histamine and its application in fish samples

Abstract:
Histamine (His) is responsible for the toxicity related to seafood consumption.However, standard detection methods do not currently meet rapid His analysis requirements. In this study, we developed an anthracene-based fluorescent sensor synthesized from the reaction of3-chloroisonicotinaldehyde and anthracen-9-ylmethylene-hydrazine. UV-Vis and fluorescence spectroscopy were used to determine the selectivity of the sensor in the existence of a range of biogenic amines. FZ-2 showed high sensitivity and selectivity towards His compared to various other biogenic amines in acetonitrile (ACN)/Acetate buffer (10 mM, pH 6.5, v/v, 1:1).The specific response to His was obtained by the substitution reaction of the primary amine of His with chlorine in the pyridine moiety of the sensor. A linear relationship was found between His concentration and fluorescence response of FZ-2 within the range 3.0-50.0 μM. The detection limit was calculated as 2.19 μM. Moreover, FZ-2 was able to monitor His in fish samples using fluorescence spectroscopy.
