Abstract:
Multistage structure optimization of natural products is an imperative approach for developing novel potential antifungal agents. Herein, various novel phthalide derivatives bearing arylhydrazine and acylthiourea skeletons were designed and synthesized according to pharmacophore-combination and homologous derivatization strategies. In vitro antifungal activity against nine phytopathogenic fungi indicated that most of the phthalide-arylhydrazine compounds exhibited excellent and broad-spectrum fungicidal activities. Among these molecules, compound 1e demonstrated superior effects on six fungi, with EC50 values ranging from 0.9 to 1.9 μg/mL. Additionally, compound 1e displayed remarkable protective and curative effects against Pyricularia oryzae, A. alternariae, Alternaria brassicae, and Valsa mali, which, respectively, infect rice, tobacco, pakchoi, and apple trees. The preliminary mechanistic investigations revealed that compound 1e exerted its antifungal effect by compromising the cell membrane permeability and effectively inhibiting spore formation and germination. Further seed germination and cytotoxicity assays indicated that compound 1e possessed favorable safety profiles and plant growth regulatory properties. These results not only provide guidance for the application of phthalide derivatives in controlling plant pathogenic fungi but also offer candidate compounds for the development of new botanical fungicides.
