Area of Science:

  • Cardiovascular Biology
  • Developmental Biology
  • Molecular Signaling

Background:

  • Axon guidance pathways like Eph/ephrin, Semaphorin, and Slit/Robo are vital for cardiac development.
  • Netrin signaling is crucial for axon guidance but its role in heart development remains unexplored.

Purpose of the Study:

  • To investigate the role of Netrin signaling in the developing murine heart.
  • To describe the novel expression pattern of Netrin-1 in embryonic mouse hearts.

Main Methods:

  • Transcriptomic analysis of embryonic mouse hearts (E8.5-E14.5).
  • RNA in situ hybridization.
  • Utilized Netrin-1Bgeo/+ reporter mice to track spatiotemporal expression.

Main Results:

  • Netrin-1 exhibits dynamic expression in the developing murine heart from E8.5 to E14.5.
  • Netrin-1 expression preferentially co-localizes with developing trabecular cardiomyocytes.
  • Highest Netrin-1 expression observed in the left ventricular trabecular myocardium, with lower levels in the right ventricle and atria.

Conclusions:

  • The identified expression pattern of Netrin-1 in the developing heart supports its potential role in cardiac development.
  • Further functional studies are required to elucidate the specific functions of Netrin signaling in the murine myocardium.
