Our research simulated the concept of oral microneedles used in oral insulin devices and worked by 1) to increase the insulin loading by layer-by-layer (LbL) loading, 2) to enhance stability by delivering the protein hormone in its solid state and 3) to prolong the release by formulating self-assembled nanoparticles (NPs) with a non-toxic anionic crosslinking agent. The core-coat nanoparticles prepared by controlled ionic gelation technique had a high surface area, facilitating the addition of multiple electrostatically attracted insulin and chitosan layers. The formation of alternative layers was evidenced by the decrement and increment in the zeta-potential value after anionic and cationic coating layers, respectively. This biodegradable multi-layered NPs matrixed with guar gum and hydroxypropyl methylcellulose was tip-loaded in microneedles. Integrating the multi-layered nanoparticles into microneedle-based oral devices could facilitate their transport and delivery through the layers of mucosa, thereby overcoming the permeation issues associated with conventional oral insulin nanoparticles. The LbL-chitosan-insulin-NPs were internalized into the cells during the cell uptake study. Sustained release was observed at pH 6.8 from the LbL-CS-INS-NPs for nearly 24 h. The intensity peaks of standard insulin were visible in the circular dichroism spectra of the stored samples, indicating protection of insulin within the layers. The minimum force required piercing through one layer of parafilm and rabbit's intestine without fracture was 0.03 N/needle. The needles formed were of good strength and high fracture force. This approach could be further utilized to deliver vaccines and hormones via any microneedle-based device for long-term drug release.
