Synthesis and Evaluation of Cleavable Thiazolidine-Modified Nucleotides for DNA Sequencing

Abstract:
A thiazolidine-modified nucleotide was synthesized for DNA sequencing by synthesis (SBS) through amidation of thiazolidine-2-carboxylic acid. The synthesized thiazolidine reversible terminator can serve as a substrate for Klenow Fragment DNA polymerase and can be incorporated into DNA strands. In addition, the thiazolidine linker can be cleaved with the assistance of palladium. After fluorescein is removed by breaking the thiazolidine linker, the next reversible terminator can be easily incorporated into the DNA.
