    Background:

    Purpose of the Study:

    • To investigate the effects of nine choline ethers on muscarinic and nicotinic receptors.
    • To elucidate the role of electronic and steric factors at the ether-oxygen in receptor stimulation.
    • To determine the potency and efficacy of these choline ethers compared to acetylcholine.

    Main Methods:

    • Isolated guinea-pig ileum longitudinal smooth muscle preparations.
    • Dose-response curve analysis in the presence of hexamethonium.
    • Assays for choline acetyltransferase and cholinesterase inhibition.
    • Receptor blockade studies using atropine.

    • Specific structural features, like beta-methyl and O-alkyl groups, may lead to secondary inhibitory pathways.
    • Choline ethers represent a class of compounds with complex interactions at cholinergic receptors.
