    Background:

    • Hepatitis B virus (HBV) infection is a global health concern.
    • Identifying reservoirs of HBV DNA is crucial for understanding viral persistence and transmission.
    • Asymptomatic carriers represent a significant population for studying HBV reservoirs.

    Purpose of the Study:

    • To investigate the distribution of hepatitis B viral DNA sequences within different blood components.
    • To identify the primary cellular reservoir of HBV DNA in asymptomatic carriers.

    Main Methods:

    • DNA hybridization techniques were employed to detect HBV DNA.
    • Cell separation methods were utilized to isolate various blood components, including plasma, platelets, polymorphonuclear leukocytes, and mononuclear cells.

    Main Results:

    • Free monomer-length hepatitis B virus DNA was abundant in the polymorphonuclear leukocyte fraction of two out of five HBsAg-positive patients.
    • Viral DNA was not detected in plasma or platelets in these patients.
    • Mononuclear cell DNA contained small amounts of a 7.2 kb unintegrated HBV genome.

    Conclusions:

    • Polymorphonuclear leukocytes appear to be the major reservoir of unit-length HBV DNA in the studied asymptomatic carriers.
    • The presence of HBV DNA in polymorphonuclear leukocytes may be due to viral replication or phagocytosis.
    • Further research is needed to elucidate the mechanisms of HBV DNA presence in these cells.
