This study used Mendelian randomization to investigate immune mediators and Parkinson's disease (PD) risk. It found causal links between 26 immune cell traits and 4 inflammatory proteins with PD, highlighting complex immune system roles.
Area of Science:
- Immunology
- Neuroscience
- Genetics
Background:
- Immune system dysfunction is correlated with Parkinson's disease (PD), but the causal link is unclear.
- Investigating immune mediators' role in PD pathogenesis is crucial for understanding disease mechanisms.
Purpose of the Study:
- To determine the causal relationship between immune cell phenotypes, circulating inflammatory proteins, and Parkinson's disease (PD) risk.
- To utilize Mendelian Randomization (MR) to analyze genetic variants associated with immune traits and PD.
Main Methods:
- Two-sample Mendelian Randomization (MR) analyses were performed.
- Genetic variants for 731 immune cell phenotypes and 91 inflammatory proteins were used as instrumental variables.
- Genome-wide association study (GWAS) data for immune traits and protein quantitative trait loci (pQTL) data for proteins were combined with PD GWAS data.
Main Results:
- A significant causal link was found between 26 immune cell traits and PD risk.
- Increased proportions of certain immune cells (e.g., resting CD4 regulatory T cells) were associated with higher PD risk.
- Alterations in other immune cell measures (e.g., FSC-A of HLA DR+CD4+ T cells) and four inflammatory proteins showed significant causal relationships with PD risk, both positive and negative.
Conclusions:
- This study establishes complex causal relationships between specific immune cell traits, inflammatory proteins, and Parkinson's disease risk.
- Immune cells and inflammation play multifaceted roles in PD development, with potential for different impacts at various disease stages.
