Biosynthesis of 2-deoxystreptamine.

K Suzukake, K Tokunaga, H Hayashi

    The Journal of Antibiotics
    |September 1, 1985
    PubMed
    Summary

Streptomyces fradiae extracts catalyze key steps in 2-deoxystreptamine biosynthesis, including amination and deamination reactions. Cell-free enzymatic synthesis of 2-deoxystreptamine from deoxyinosose was successfully demonstrated.

    Area of Science:

    • Biochemistry
    • Microbiology
    • Enzymology

    Background:

    • Neomycin, an essential antibiotic, is synthesized via the 2-deoxystreptamine pathway.
    • Understanding the enzymatic steps is crucial for antibiotic biosynthesis research.

    Purpose of the Study:

    • To elucidate the enzymatic reactions involved in 2-deoxystreptamine biosynthesis by Streptomyces fradiae.

    Main Methods:

    • Enzyme assays using cell-free extracts of Streptomyces fradiae.
    • Ion-exchange chromatography (DEAE-Sepharose CL-6B) and gel filtration chromatography (Sepharose CL-6B) for enzyme purification and molecular weight estimation.

    Main Results:

    • Identified three key enzymatic activities: deoxyinosose amination, deoxyinosamine to 2-deoxystreptamine conversion, and 2-deoxystreptamine deamination.
    • Glutamine served as an effective amino donor; NAD+ stimulated deoxyinosamine conversion.
    • Purified an aminotransferase (130,000 daltons) and a deoxyinosamine dehydrogenase.

    Conclusions:

    • Enzymatic synthesis of 2-deoxystreptamine from deoxyinosose is feasible using cell-free extracts.
    • Characterized key enzymes in the 2-deoxystreptamine biosynthetic pathway, providing insights into neomycin production.
