Cell luminescence assays can be affected by the moon phase. This study found that autoactivation in HeLa 9903 cells correlates with increased moonlight, suggesting a novel environmental factor influencing in vitro assays.
Area of Science:
- Cell biology
- Environmental science
- Biochemistry
Background:
- The HeLa 9903 cell line is utilized in the in vitro endocrine disruption assay (OECD 455).
- This assay measures luminescence changes in response to estrogen receptor agonists.
- Autoactivation, or spontaneous luminescence increase, has been observed in this test system.
Purpose of the Study:
- To investigate the underlying cause of autoactivation in the HeLa 9903 cell line.
- To determine if external environmental factors influence the assay's baseline luminescence.
Main Methods:
- HeLa 9903 cells were cultured and seeded under varying moon phases.
- Luminescence was measured the subsequent day.
- Enzyme-linked immunosorbent assay (ELISA) was employed to detect 17β-estradiol expression.
Main Results:
- A statistically significant correlation was found between the phase of the moon and increased cell luminescence.
- Autoactivation was more probable when moonlight visibility exceeded approximately 87.7% (full moon).
- ELISA results indicated that this autoactivation was ligand-independent, not due to endogenous 17β-estradiol production.
Conclusions:
- The phase of the moon is a significant factor influencing the autoactivation of HeLa 9903 cells in vitro.
- This finding represents the first documented relationship between lunar cycles and cell activation in cultured systems.
- Results necessitate careful consideration of lunar phases when planning and conducting studies using this endocrine disruption assay.
