Abstract:
This study investigated the remodeling process of the cell wall in the postharvest fruiting body of Coprinus comatus. We monitored the changes in the content of cell wall components, the structure of cell wall polysaccharides, and the expression of key carbohydrate-active enzymes (CAZymes) involved in cell wall metabolism during the storage period. The results demonstrated a decreasing trend in the structural components (glucan and chitin) of the cell wall, accompanied by a significantly compromised structure. The cell wall polysaccharides were fractionated, revealing that the polysaccharides located in the outermost layer were predominantly degraded during storage. This depolymerization resulted in significant changes in molecular weight distribution and monosaccharide composition. NMR analyses indicated that the configuration and linkage patterns of glycosidic bonds also underwent substantial alterations. Furthermore, qRT-PCR validation showed that GH5, GH27, CE9, AA3_2, and CBM5 potentially promoted cell wall degradation, while GT2, GT41, and CBM13 facilitated cell wall synthesis.
