Abstract:
Pulmonary arterial hypertension (PAH) is a progressive and life-threatening disorder characterized by elevated pulmonary vascular resistance. Mutations in the bone morphogenetic protein receptor type 2 (BMPR2) gene are strongly implicated in its pathogenesis. This review systematically examines the impact of BMPR2 mutations on the BMP/TGF-β signaling pathway and the subsequent pathological processes, including dysregulated cellular proliferation, apoptosis resistance, inflammatory activation, and metabolic reprogramming. Furthermore, potential therapeutic strategies targeting the BMPR2 pathway are discussed. The review also explores the "second hit" hypothesis and the modulatory role of estrogen in PAH, providing a theoretical framework for the development of precision therapies.
