A novel ruthenium(II) complex (complex I) effectively cleaves DNA and shows potent antibacterial activity against Staphylococcus strains. Its phenanthroline ligand enhances lipophilicity, boosting its antimicrobial efficacy compared to a similar bipyridine complex.
Area of Science:
- Coordination Chemistry
- Photochemistry
- Antimicrobial Agents
Background:
- Ruthenium(II) complexes are explored for their diverse chemical and biological properties.
- Understanding structure-activity relationships is key for developing novel therapeutic agents.
Purpose of the Study:
- To synthesize and characterize a novel ruthenium(II) compound, cis-[Ru(phen)2(4-bzpy)(Cl)](PF6) (complex I).
- To investigate its photochemical behavior, DNA cleavage ability, and antimicrobial activity.
- To compare its efficacy with a related ruthenium(II) complex (complex II).
Main Methods:
- Synthesis and full characterization (electrochemical, spectroscopic, X-ray diffraction).
- Density Functional Theory (DFT) calculations.
- Photochemical studies (blue light irradiation).
- DNA cleavage assays.
- Antimicrobial activity testing against bacterial strains.
Main Results:
- Complex I was successfully synthesized and characterized.
- Blue light irradiation of complex I induced 4-benzoylpyridine ligand release and generated reactive oxygen species.
- Complex I demonstrated significant DNA cleavage ability.
- Both complexes I and II exhibited antibacterial activity against Staphylococcus aureus and Staphylococcus epidermidis.
- Complex I showed superior antibacterial activity compared to complex II.
Conclusions:
- The synthesized ruthenium(II) complex exhibits promising photochemical and DNA-cleaving properties.
- Complex I possesses significant antimicrobial activity, with the phenanthroline ligand playing a crucial role in enhancing its efficacy.
- Ruthenium(II) complexes represent a potential avenue for developing new antimicrobial agents.
