Exploring RNA-protein interactions is crucial for understanding the operational modes of both constituents of RNA-protein complexes. Here, we present a protocol for brain tissue processing and its application in studying circular RNA (circRNA)-protein interactions. We describe steps for cryogenic biopulverization of the tissue and UV irradiation, followed by a circRNA pull-down protocol. The circRNA pull-down procedure described here represents a novel approach for exploring the circRNA interactome in vivo.
