Personalized compounded medications for companion animals

Abstract:
The growing demand for personalized veterinary therapies highlights a critical need for dosage forms tailored to the anatomical and physiological variability among companion animals. Yet, the limited availability of weight-adjusted, palatable and flexible formulations often forces veterinarians to rely on off-label human medicines or manual compounding, introducing risks of imprecise dosing and excipient toxicity. To address this gap, we present an approach to on-demand veterinary compounding via semi-solid extrusion (SSE) 3D printing, using peanut butter, a species-preferred, drug-compatible excipient, as the primary matrix. The formulation was loaded with prednisolone (PRE) by compounding crushed Prezolon® tablets into a starch-enhanced matrix, producing chewable units with tunable doses. The printed chewable tablets exhibited robust extrusion characteristics, excellent dose linearity (R2 > 0.99), immediate drug release and mechanical properties conducive to voluntary oral intake. Stability over 14 days under refrigerated storage confirmed their suitability for veterinary clinic preparation and at-home administration. This work introduces a scalable compounding platform to improve pharmacological care for companion animals, particularly those requiring low dose, chronically administered medications.
