Serial adaptation of viruses to inhibitors enhances the understanding of the mechanism of inhibition, helps to identify physical target sites of compounds, and serves as a test for the genetic barrier to reducing virus susceptibility. Virus adaptations can be carried out in vivo or in tissue culture using sub-inhibitory concentrations of compounds that are increased with each passage of the virus or kept static. Here we describe the method of serially adapting a virus in tissue culture.
