Abstract:
Conventional fluorescence-guided oncotherapy has high fluorescence background due to in situ excitation, which limits its clinical applications. Near infrared (NIR)-emitting persistent luminescent nanoparticles possess distinctive advantages, such as the ability to function without in-situ excitation, a high signal-to-noise ratio, and deep tissue penetration. These properties make them highly promising for in vivo bioimaging applications. Here, we develop an innovative nanoplatform for imaging-guided gene and optothermal combination therapy, achieving highly efficient tumor cell eradication. This system loads the Bax gene and indocyanine green (ICG) into mesoporous silica nanoparticles, which are encapsulated by ZnGa2O4:Cr3+ (ZGC). This multifunctional design enables simultaneous NIR-persistent luminescent imaging and synergistic cancer treatment. The results revealed that the nanoplatform achieved an excellent combined therapeutic effects, effectively inhibiting solid tumors through simultaneous gene therapy and photothermal treatment under 808 nm NIR laser irradiation in mice. These data provide the promising potential of this nanocomposite system to build a multi-modal therapeutic platform under persistent luminescent imaging-guided visual treatment of tumor.
