Purpose of the Study:

  • To evaluate the efficacy and safety of Olezarsen in patients with moderate or severe hypertriglyceridemia.
  • To assess the placebo-adjusted change in triglyceride levels at 6 months.

Main Methods:

  • Phase 3, international, double-blind, randomized, placebo-controlled trial.
  • 1349 patients with moderate or severe hypertriglyceridemia received monthly subcutaneous Olezarsen (50-mg or 80-mg) or placebo.
  • Primary outcome: least-squares mean percent change in triglyceride level from baseline to 6 months.

Main Results:

  • Olezarsen significantly reduced triglyceride levels compared to placebo.
  • The 50-mg dose showed a placebo-adjusted reduction of -58.4 percentage points (P<0.001).
  • The 80-mg dose showed a placebo-adjusted reduction of -60.6 percentage points (P<0.001).

Conclusions:

  • Olezarsen treatment led to a significantly greater reduction in triglyceride levels than placebo in patients with moderate hypertriglyceridemia and elevated cardiovascular risk.
  • The incidence of serious adverse events was similar across treatment groups.
  • Olezarsen represents a promising therapeutic option for managing hypertriglyceridemia.
