Background:

Purpose of the Study:

  • To investigate bidirectional causal relationships between thyroid diseases (hypothyroidism, hyperthyroidism, Hashimoto thyroiditis, Graves disease) and psoriasis (PsO) and its subtypes (psoriasis vulgaris (PV), psoriatic arthritis (PsA)).

Main Methods:

  • Conducted a bidirectional two-sample Mendelian randomization (MR) analysis using GWAS summary statistics from European populations.
  • Selected independent single nucleotide polymorphisms (SNPs) as instrumental variables.
  • Employed MR-Egger, inverse-variance weighted (IVW), and weighted median methods, with MR-PRESSO for outlier detection.

Main Results:

  • Hypothyroidism showed a suggestive association with increased PsO risk (OR=1.059) and a significant association with PsA risk (OR=1.184).
  • Psoriasis vulgaris (PV) was suggestively associated with increased risk of Graves disease (GD) (OR=1.113) and hyperthyroidism (OR=1.053).
  • PV was associated with a decreased risk of hypothyroidism (OR=0.977).

Conclusions:

  • Presents genetic evidence supporting bidirectional causal relationships between thyroid diseases and psoriasis.
  • Findings suggest shared inflammatory pathways underlying these comorbidities.
