    Background:

    • Erythrocytes, or red blood cells, possess varying amounts of N-acetylneuraminic acid (sialic acid) on their surface.
    • Neuraminidase is an enzyme that cleaves sialic acid residues from cell surfaces.
    • The role of sialic acid in erythrocyte immunogenicity is not fully understood.

    Purpose of the Study:

    • To investigate the influence of neuraminidase on the immunogenicity of heterologous erythrocytes in mice.
    • To determine if sialic acid acts as an intrinsic immunological adjuvant or antigenic determinant.

    Main Methods:

    • Sheep erythrocytes (high sialic acid) and rabbit erythrocytes (low sialic acid) were treated with neuraminidase.
    • Immunogenicity was assessed by serum haemagglutination titres in mice.
    • Experiments included the use of extrinsic adjuvants and complement-deficient mice.

    Main Results:

    • Neuraminidase treatment caused a ten-fold reduction in the immunogenicity of sheep erythrocytes.
    • Rabbit erythrocytes showed resistance to neuraminidase treatment.
    • Sialic acid demonstrated intrinsic adjuvant activity, dependent on complement component C5 and T-cells.

    Conclusions:
