Palbociclib monotherapy demonstrated disease control in over half of advanced liposarcoma patients, but showed modest activity. Further research into biomarker-driven and combination therapies is warranted for improved outcomes in well-differentiated (WDLPS) and dedifferentiated liposarcomas (DDLPS).
Area of Science:
- Oncology
- Medical Science
- Pharmacology
Background:
- Well-differentiated liposarcoma (WDLPS) and dedifferentiated liposarcoma (DDLPS) are distinct subtypes often driven by CDK4 amplification.
- CDK4/6 inhibitors like palbociclib show therapeutic promise, but real-world data for liposarcoma treatment is limited.
Purpose of the Study:
- To evaluate the real-world effectiveness and safety of palbociclib monotherapy in patients with advanced WDLPS or DDLPS.
- To assess progression-free survival (PFS), overall survival (OS), response rates, and adverse events associated with palbociclib treatment.
Main Methods:
- Retrospective analysis of 21 patients with advanced WDLPS or DDLPS treated with palbociclib monotherapy.
- Data collected from 16 Turkish Oncology Group centers between 2019 and 2022.
- Outcomes assessed included PFS, OS, objective response rate (ORR), disease control rate (DCR), and safety profile.
Main Results:
- Median PFS was 5.3 months and median OS was 9.1 months.
- No objective responses were observed (ORR 0%), but disease control was achieved in 57.1% of patients.
- Common adverse events included anemia (52.4%) and neutropenia (33.3%); 71.4% experienced some adverse events.
Conclusions:
- Palbociclib monotherapy exhibited modest activity, with disease stabilization observed in a subset of patients with advanced liposarcoma.
- The findings underscore the need for biomarker-driven approaches and combination strategies to enhance therapeutic efficacy in WDLPS and DDLPS.

Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...
