Storage-induced spectral changes of human platelet-rich plasma revealed by SERS

Abstract:
The article presents the results of an analysis of Raman spectra from platelet-rich plasma from a healthy volunteer over a period of one working day (6 h), one week, and up to six weeks, storing at +4 °C. The data were processed and analyzed using principal component analysis, ANOVA analysis and linear discriminant analysis. Spectral markers of Raman spectroscopy: 1002 cm-1, 1155 cm-1, 1445 cm-1, 1517 cm-1, 1654 cm-1 corresponding with proteins, lipids, amides and amino acids were revealed and selected for analysis. It was found that spectral changes in the platelet spectrum begin three hours after the start of the study and continue throughout the six-week period. It was revealed that the time limit for studying human platelets using Raman spectroscopy without significant changes was up to four hours after sampling the platelet-rich plasma.
