Methods for the synthesis of natural UDP-sugars and synthetic analogues

Abstract:
This review highlights recent advancements in enzymatic and chemoenzymatic methods for the synthesis of natural and unnatural UDP-sugars, essential donors in glycosylation processes for the formation of polysaccharides like starch and glycogen, lipopolysaccharides, and glycosphingolipids. The most commonly synthesized nucleotide sugars include uridine 5'-diphosphate-glucose (UDP-Glc), UDP-galactose (UDP-Gal), UDP-xylose (UDP-Xyl), UDP-N-acetylglucosamine (UDP-GlcNAc), and UDP-N-acetylgalactosamine (UDP-GalNAc), along with numerous synthetic analogues. In order to create UDP-sugars, various techniques have been investigated so far. One-pot multi-enzyme systems enhance efficiency through cofactor regeneration and enzyme reuse, reducing purification. Recombinant expression in Escherichia coli and Saccharomyces cerevisiae, etc., enables scalable in vitro and in vivo synthesis. Advances in enzyme engineering, including deep learning-guided design, have improved stability and broadened substrate scope. By using chemical synthesis in combination with enzymatic methods, a wide range of natural and synthetic UDP-sugars can be produced. The synthesis of numerous natural and synthetic UDP-sugar analogues is comprehensively discussed in this review.
