  • Bipolar disorder (BD) is associated with observed changes in brain and blood transcriptomes.

Purpose of the Study:

  • To investigate if blood transcriptome changes in bipolar disorder (BD) reflect corresponding alterations in the cortical transcriptome.
  • To identify potential diagnostic or theranostic markers for BD within the blood transcriptome.

Main Methods:

  • RNA levels were quantified in blood samples from 19 individuals with BD and 19 healthy controls using Affymetrix Human Exon 1.0 ST Arrays.
  • Statistical analysis identified significant differences in RNA levels between groups, followed by pathway analysis using the Panther Classification System.

Main Results:

  • A total of 19,473 coding and non-coding RNAs were detected.
  • Significant alterations in 98 RNAs (65 with increased levels) were observed in individuals with BD compared to controls (fold ≥±20%, p < 0.05).
  • Affected pathways included toll-like receptor and Wnt signaling, viral response, cell-cell signaling, protein metabolism, and synaptic organization. Four altered RNAs in blood were consistent with previous cortical findings.

Conclusions:

  • Blood transcriptome alterations in BD impact peripheral biochemical homeostasis and show overlap with cortical changes.
  • These findings support the potential of blood transcriptome analysis for identifying diagnostic or theranostic markers in bipolar disorder.
