Summary

Patient-derived organoids (PDOs) accurately predict treatment response in metastatic colorectal cancer (mCRC). This study identified predictors for successful PDO establishment and validated their utility in forecasting patient outcomes with 5-fluorouracil and oxaliplatin therapies.

Background:

  • Accurate prediction of treatment response in metastatic colorectal cancer (mCRC) is crucial for optimizing patient care and minimizing toxicity.
  • Patient-derived organoids (PDOs) offer a promising preclinical model for predicting therapeutic efficacy, but require further validation in prospective studies.

Purpose of the Study:

  • To identify predictors for successful establishment of PDOs from mCRC patients.
  • To evaluate the predictive value of PDOs for patient response to systemic treatment.
  • To assess the association between PDO drug sensitivity and patient survival outcomes.

Main Methods:

  • Metastatic biopsies were obtained from mCRC patients prior to new systemic treatment for PDO establishment.
  • PDOs were cultured and drug sensitivity was assessed using a seven-drug panel.

  • Predictors for successful mCRC PDO establishment were identified.
  • PDOs demonstrated significant potential in accurately predicting patient outcomes during systemic therapy.
  • The study validates the use of PDOs, particularly with 5-fluorouracil and oxaliplatin, for treatment response prediction in mCRC.
