Nociception01:44

Nociception

Nociception—the ability to feel pain—is essential for an organism’s survival and overall well-being. Noxious stimuli such as piercing pain from a sharp object, heat from an open flame, or contact with corrosive chemicals are first detected by sensory receptors, called nociceptors, located on nerve endings. Nociceptors express ion channels that convert noxious stimuli into electrical signals. When these signals reach the brain via sensory neurons, they are perceived as pain. Thus, pain helps the...

Pathophysiology of Peptic Ulcer Disease: Injurious Factors

Peptic ulcers are sores on the stomach's inner lining and the upper small intestine, which are the result of disruptions in the mucosal layer that houses parietal cells which produce gastric acid, and chief cells which secrete pepsinogen.
In the antrum region, G cells secrete the gastrin hormone that binds to gastrin-cholecystokinin-B (CCK2) receptors on parietal and enterochromaffin-like (ECL) cells in the fundic glands. Simultaneously, the vagus nerve releases acetylcholine, which binds to M3...
Determinants of Bacterial Pathogenicity and Virulence01:20

Determinants of Bacterial Pathogenicity and Virulence

Pathogenic bacteria employ a variety of strategies to establish infections, including the secretion of extracellular enzymes that act as potent virulence factors. These enzymes facilitate bacterial colonization of host tissues and help evade immune surveillance. By targeting structural components of host tissues and interfering with immune mechanisms, these enzymes play a pivotal role in disease progression.Extracellular Enzymes Facilitating Tissue Invasion: Several bacterial pathogens secrete...

Summary

Polydatin treatment reduced inflammatory cytokines like IL-6 and TNF-α in dental pulp stem cells stimulated by lipoteichoic acid (LTA). This natural compound also increased IL-10 levels, suggesting a role in managing dental caries inflammation.

Area of Science:

  • Oral Biology
  • Immunology
  • Pharmacology

Background:

  • Dental caries initiation involves Gram-positive bacteria and lipoteichoic acid (LTA) binding to dental pulp cells.
  • This interaction triggers immune responses, inflammation, and potential pulp necrosis.
  • Polyphenolic compounds, such as polydatin, show promise in modulating inflammatory mediators for healing.

Purpose of the Study:

  • To evaluate the effect of polydatin on cytokine expression in lipoteichoic acid (LTA)-stimulated human dental-pulp stem cells (hDPSCs).
  • To assess the impact of varying polydatin concentrations on key inflammatory markers.

Main Methods:

  • Human dental-pulp stem cells (hDPSCs) were stimulated with LTA and subsequently treated with polydatin (0.01 µM, 0.1 µM, 1 µM).
  • Cytokine levels (IL-6, IL-8, IL-10, TNF-α) were quantified using RT-qPCR and ELISA.

Main Results:

  • Polydatin significantly decreased Interleukin-6 (IL-6) and Tumor Necrosis Factor-alpha (TNF-α) levels.
  • Reduced Interleukin-8 (IL-8) mRNA and protein levels were observed with specific polydatin concentrations.
  • Interleukin-10 (IL-10) levels were significantly increased across all tested polydatin concentrations.

Conclusions:

  • Polydatin effectively modulates inflammatory cytokine production in LTA-stimulated hDPSCs.
  • The compound suppresses pro-inflammatory cytokines (IL-6, IL-8, TNF-α) and enhances the anti-inflammatory cytokine IL-10.
  • Polydatin demonstrates potential therapeutic benefits for managing inflammation associated with dental caries.
