Abstract:
Macromolecular crowding, the dense packing of macromolecules within cells, significantly influences enzyme kinetics, cell signaling, liquid-liquid phase separation, and disease progression. This review explores the multifaceted effects of crowding on osmotic balance, intracellular compartmentalization, and metabolic efficiency. We highlight the exclusion volume effect's role in protein folding, stability, and osmotic homeostasis. Crowding also modulates enzyme activity and kinetics, contributing to metabolic regulation, and plays a crucial role in forming membraneless organelles. Dysregulation of macromolecular crowding is implicated in neurodegenerative diseases, cancer, and type 2 diabetes, highlighting its importance for therapeutic development. Future research should focus on elucidating crowding mechanisms, developing in vivo models, and exploring therapeutic interventions targeting macromolecular crowding.
