Background:

Purpose of the Study:

  • To evaluate the antimicrobial efficacy of ExAFs derived from native LAB against MDR *Escherichia coli* strain L1PEag1.
  • To characterize the structural damage induced by ExAFs on bacterial cells.
  • To identify bioactive metabolites within the most effective ExAF using LC-MS/MS.

Main Methods:

  • Screening of 14 ExAFs using time-kill assays.
  • Assessment of bacterial ultrastructural damage via scanning and transmission electron microscopy (SEM/TEM).
  • Metabolomic profiling of the most potent ExAF (E10) using LC-MS/MS with SWATH acquisition.

  • ExAF E10, from *Weissella cibaria* UTNGt21O, showed the strongest inhibition (17.12 ± 0.22 mm zone of inhibition).

  • SEM/TEM revealed membrane disruption and intracellular damage, indicating a membrane-targeting mechanism.
  • Metabolomic analysis of E10 identified 22 bioactive compounds, including peptides and flavonoids, with similarities to RiPPs.

Conclusions:

  • ExAFs, particularly E10, are potent, peptide-rich, bio-based antimicrobial candidates.
  • The presence of RiPP analogs suggests multifaceted bactericidal effects.
  • ExAFs represent promising eco-friendly alternatives for managing MDR pathogens in food safety and therapeutics.
