Ultraviolet irradiation of 1-aryltriazenes with azidotrimethylsilane generates hydrazoic acid for efficient ipso-azidation. This method successfully converts various 1-aryltriazenes, even on proteins, into aryl azides.
Area of Science:
- Organic Chemistry
- Photochemistry
- Synthetic Methodology
Background:
- 1-aryltriazenes are versatile precursors in organic synthesis.
- Azidotrimethylsilane is a common reagent for introducing azide groups.
- Ultraviolet (UV) irradiation is increasingly used to drive chemical transformations.
Purpose of the Study:
- To develop an efficient method for ipso-azidation of 1-aryltriazenes.
- To investigate the mechanism of UV-promoted azidation.
- To demonstrate the functional group tolerance and applicability of the reaction.
Main Methods:
- Photochemical reaction of 1-aryltriazenes with azidotrimethylsilane in methanol under UV irradiation (λmax = 370 nm).
- Optimization of reaction parameters (solvent, concentration, irradiation time).
- Mechanistic studies involving in situ generation of hydrazoic acid.
Main Results:
- UV irradiation efficiently promoted ipso-azidation of 1-aryltriazenes.
- In situ generated hydrazoic acid was identified as the key intermediate for the transformation.
- The reaction exhibited high functional group tolerance, enabling the conversion of diverse substrates.
- The method was successfully applied to convert 1-aryltriazenes on a protein to aryl azides.
Conclusions:
- UV-promoted ipso-azidation of 1-aryltriazenes using azidotrimethylsilane is a viable and efficient synthetic route.
- The reaction proceeds via in situ generated hydrazoic acid and tolerates a wide range of functional groups.
- This methodology offers a valuable tool for introducing aryl azides, including in bioconjugation applications.

Azide ions act as good nucleophiles and react with unhindered alkyl halides to form alkyl azides. Alkyl azides do not participate in further nucleophilic substitution reactions, thereby eliminating the chances of polyalkylated products.
